닥나무 잎 에탄올 추출물의 프리바이오틱스 특성 및 H2O2와 LPS로 유도된 산화 스트레스와 염증에 대한 HT-29 세포 보호 효과

This study investigated the antioxidant activity of a 20% ethanolic extract from Broussonetia kazinoki leaves (EBK), its protective effects against lipopolysaccharide (LPS)- and H2O2-induced cytotoxicity in HT-29 cells, and its potential prebiotic activity in promoting probiotic growth. EBK exhibited high total phenolic and flavonoid contents and showed strong antioxidant capacity, as determined by 2,2'-azino-bis (3-ethyl benzothiazoline-6-sulfonic acid) (ABTS), 2,2-diphenyl- 1-picrylhydrazyl (DPPH) radical scavenging assays, and ferric reducing/antioxidant power. EBK significantly enhanced the viability and titratable acidity of the probiotic strains Limosilactobacillus reuteri and Levilactobacillus brevis. In HT-29 cells, EBK increased cell viability and suppressed intracellular reactive oxygen species (ROS) generation, thereby alleviating LPS- and H2O2-induced cytotoxicity. Furthermore, EBK markedly downregulated the expression of inflammatory signaling proteins, including Toll-like receptor-4 (TLR-4), myeloid differentiation primary response 88 (MyD88), phosphorylated NF-κB (p-NF-κB), and inducible nitric oxide synthase (iNOS) in LPS-stimulated HT-29 cells. High-performance liquid chromatography (HPLC) identified chlorogenic acid and rutin as the major bioactive compounds in EBK. These findings suggested that EBK possessed potent antioxidant, anti-inflammatory, and prebiotic properties, highlighting its potential application as a functional food ingredient.
