This study investigated the neuroprotective effects of fermented Protaetia brevitarsis larval hydrolysate (FPB) in mice fed with a high-fat diet. FPB administration improved memory deficits, as evidenced by the performance of mice in the Y-maze, passive avoidance, and Morris water maze tests. FPB significantly protected the antioxidant systems of mice by regulating malondialdehyde, superoxide dismutase, and reduced glutathione levels in brain tissues. In addition, FPB reversed cerebral mitochondrial dysfunction by modulating reactive oxygen species levels, mitochondrial membrane potential, and adenosine triphosphate content. FPB also attenuated the inflammatory response by down-regulating the Tolllike receptor-4/nuclear factor-kappa B pathway in whole brain tissues. Moreover, FPB mitigated amyloid β and tau protein accumulation by modulating insulin-degrading enzyme and the phosphoinositide 3-kinases/protein kinase B pathway in the hippocampus. Notably, FPB suppressed neurotoxicity by modulating apoptosis-related protein expressions. In conclusion, these results revealed that FPB can improve cognitive impairment through oxidative stress, neuroinflammation, and neurotoxicity.
