Abstract
Saccharina japonica is a type of brown algae commonly used in Asian countries. Brown algae contain large amounts of alginic acids; evaluating its colorimetric properties is crucial for determining its quality. The ultrasonic-assisted extraction technique (UAT) was performed on S. japonica to extract high-quality alginic acid, followed by activated carbon treatment (UATC) and ethanol treatment (UATE). Chromaticity was evaluated using the Hunter color system to assess the effectiveness of UATE in pigment removal. The extraction yields were 30% for UAT and 23% for UATC and UATE. The polysaccharide content was analyzed as follows: 41.579±0.16 mg% for UAT, 45.459±0.47 mg% for UATC, and 42.174±0.17 mg% for UATE. Antioxidant activity assays were performed to assess 2,2'-azino-bis(3-ethylbenzothiazoline- 6-sulfonic acid), 2,2-diphenyl-1-picrylhydrazyl, and H2O2 scavenging activities; all increased in a concentration-dependent manner. The total polyphenol content was quantified as 2.37±0.12 mg/g for UAT, 0.86±0.18 mg/g for UATC, and 0.09±0.01 mg/g for UATE. These results suggest that pigment removal using ethanol is highly effective for efficacy screening and provides fundamental data useful for application in the food, cosmetic, and pharmaceutical industries.
