- Abstract
- A protocol for in vitro propagation of variegated Solomon's seal (Polygonatum odoratum Druce var. pluriflorum Ohwi f. variegatum Y. N. Lee) was developed. Rhizome explants were cultured on Anderson's basal nutrient (AM) medium supplemented with different concentrations and combinations of plant growth regulators. Adventitious shoots were induced directly on the surface of explants when the culture medium was supplemented with cytokinin alone. The highest frequency of shoot induction (100%) was obtained when the explants were cultured on AM medium supplemented with 1.0 mg l(-1)2,4-D, 2.0 mg l(-1) 2iP, and 0.5 mg l(-1) TDZ, with a mean number of 7.6 shoots. The greatest frequency of rooting (100%), with maximum mean number of 5.2 roots were obtained on half strength AM medium supplemented with 1.0 mg l(-1) NAA. The survival rate of regenerated plants transferred to the greenhouse was 80.8 +/- 1.2%. The acclimatized plants grew normally without morphological abnormalities or variations.
