Abstract Barnacles are sessile marine crustaceans with significant ecological and economic impacts, particularly due to their widespread distribution and strong substrate adhesion. Understanding their reproductive physiology is crucial for managing their effects on marine industries and aquaculture. This review examines optimized protocols for tissue sample processing in barnacle reproductive studies, focusing on species native to Korean coastal waters such as Amphibalanus eburneus and Balanus trigonus. Key steps, including sample collection, decalcification, fixation, tissue dissection, processing, embedding, sectioning, staining, and mounting, are detailed, with an emphasis on minimizing tissue damage and maximizing histological clarity. Comparative analysis of decalcification agents, such as commercial Decalcifier and acetic acid–ethanol mixtures, reveals differences in tissue preservation and processing efficiency. The review also discusses challenges unique to barnacle histology, including the removal of calcareous exoskeleton and the preparation of high-quality sections for microscopic evaluation of reproductive structures. The summarized protocols and recommendations aim to enhance reproducibility and reliability in barnacle reproductive biology research, providing a foundation for further studies on their biology and management in marine environments.
