Formulation Development of Ketoprofen Microemulsion Gel

초록

Ketoprofen is a widely used nonsteroidal anti-inflammatory drug (NSAID) but has low oral bioavailability and gastrointestinal side effects, which limit its therapeutic efficacy. This study aimed to develop a microemulsion-based gel formulation to enhance topical delivery while improving spreadability and reducing stickiness compared to commercial products. Oleic acid, Tween 80, and Transcutol P were utilized as the oil phase, surfactant, and cosurfactant, respectively. A pseudo-ternary phase diagram showed that a Smix ratio of 2:1 was optimal for stable microemulsion formation. The selected microemulsion was characterized for particle size, polydispersity index (PDI), and zeta potential and incorporated into gels containing Carbopol 934 at concentrations of 0.5%, 1.0%, and 1.5%. Of the formulations identified as F1-F3, F2 (1.0% Carbopol 934) exhibited optimal physicochemical properties, viz., viscosity, stickiness, and gel strength, and was comparable to Rheuma Gel. The developed formulation offers a potential means of improving topical ketoprofen delivery. Further, in vitro and ex vivo studies are required to confirm the feasibility of the formulation for clinical applications and commercialization.
