This study established the novel molecular marker system for identifying the endangered Asiatic black bears (Ursus thibetanus ussuricus)being restored in South Korea. Using whole genome sequence information, a total of twenty microsatellite (MS) markers were newlydeveloped after genetic marker evaluation by multiplex polymerase chain reaction (PCR) mediated genotyping. The number of allelesof twenty MS markers ranged from 3 to 10, the average number was 6.05 alleles. The mean values of observed heterozygosity (Hobs), expected heterozygosity (Hexp), and polymorphism information content (PIC) values were 0.683, 0.715, 0.658 from the thirty-threeintroduced animals, respectively. The Fis was –0.385 to 0.438, average 0.052. The probability of identical individuals (PI) levels arisingunder random mating (PIrandom), half-sib mating (PIhalf-sib), and full-sib mating (PIfull-sib) were 1.58×10-18, 2.79×10-14, and 4.75×10-⁸,respectively. The results of this study showed that this new MS marker system had the high genetic diversity levels and the quitelylow PI levels, suggesting that this marker system may be useful to identify the newborn cubs found at the wild and to seek for theirparents of the wild-born cubs. This molecular marker system will contribute to managing bear population and improving genetic diversityin the restoring Asiatic black bears in South Korea
