First report of Zoysia japonica leaf spot disease caused by Didymella glomerata in Korea

초록

Leaf spot disease poses a persistent and substantial threat to the health and aesthetic quality of turfgrass worldwide, including that commonly cultivated in golf courses and green areas across Korea. We investigated the causal agent of an unidentified leaf spot disease characterized by distinct necrotic lesions on Zoysia japonica. The pathogen was isolated from symptomatic tissues and identified using morphological and molecular analyses. Morphological characterization was conducted by culturing the pathogen on potato dextrose agar to examine colony morphology, followed by microscopic examinations. The pathogen produced brown, globose pycnidia and hyaline conidia. Molecular identification was performed using PCR amplification and sequencing of the internal transcribed spacer region. Sequence analysis revealed that the isolate exhibited phylogenetic identity with Didymella glomerata reference strains. This study provides the first report of Zoysia grass leaf spot disease caused by D. glomerata in Korea.
