H2O2 및 ethanol로 유도된 간세포 독성에 대한 엉겅퀴(Cirsium japonicum var. maackii) 잎 추출물의 보호 효과

This investigation was designed to evaluate the antioxidant and hepatoprotective properties of a 40% ethanolic extract of C. japonicum var. maackii leaves (CJL). High-performance liquid chromatography (HPLC) analysis identified chlorogenic acid (27.36 μg/mg) as a major bioactive compound in CJL. Based on its phytochemical profile, CJL showed considerable antioxidant capacity, with total phenolic content of 68.58 mg GAE/g and total flavonoid content of 47.30 mg RE/g. It was further supported by considerable 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activities, ferric reducing antioxidant power (FRAP), and inhibition of malondialdehyde (MDA) production. In terms of metabolic regulation, CJL showed inhibitory effects on α-glucosidase activity and on the formation of advanced glycation end-products (AGEs), while concentration-dependently enhancing the activities of alcohol-metabolizing enzymes, including alcohol dehydrogenase (ADH) and aldehyde dehydrogenase (ALDH). Furthermore, HepG2 cell experiments showed that CJL increased viability and lowered intracellular reactive oxygen species (ROS) levels in response to H2O2 and ethanol exposure. These findings suggest that CJL possesses antioxidant, anti-glycation, and alcohol-metabolizing activities that may collectively contribute to protection against alcohol- and oxidative stress-induced hepatocyte injury in vitro. Taken together, these results support the potential of CJL as a candidate natural material for the management of alcoholic liver disease.
