This study investigated gentamicin (GEN) residues in the edible tissues of pigs following oral administration and identified the appropriate withdrawal time (WT) of GEN in porcine edible tissues. Forty-two healthy pigs were orally administered GEN at doses of 1.1 mg/kg body weight (BW) (GEN-1, n=20) and 2.2 mg/kg BW (GEN-2, n=20) once daily for three consecutive days. After drug administration, tissue samples were collected from four pigs at 1, 3, 5, 7, and 14 d post- treatment. The GEN residue concentrations in porcine tissues were determined using liquid chromatography-tandem mass spectrometry (LC-MS/MS). The correlation coefficient values of the calibration curve ranged from 0.9961 to 0.9996, and the limits of detection (LOD) and quantification (LOQ) were 0.003-0.008 mg/kg and 0.01-0.025 mg/kg, respectively. Recoveries in the muscle, liver, kidney, and fat tissues were 55.51-73.15%, 69.65-78.21%, 66.56-83.29%, and 96.76-107.74%, respectively, with coefficients of variation of less than 11.16%. In both the GEN-1 and GEN-2 groups, GEN concentrations in the muscle, liver, and fat samples were below the LOQ on day 1 post-treatment. However, in both groups, GEN was detected in the kidney samples at levels above the LOQ up to day 7 after drug administration. According to the guidelines of the European Medicines Agency for the determination of withdrawal periods for edible tissues, the withdrawal periods for GEN-1 and GEN-2 in porcine tissues were established as two and nine days, respectively. In conclusion, the developed analytical method is suitable for detecting GEN in porcine tissues, and the estimated WT of GEN can contribute to the safety of porcine products.
