Simultaneous UPLC-MS/MS Analysis and Antioxidant Activity Evaluation of Lonicera japonica Flos Extracts Prepared by warm water and Hydrolyzed Extraction

Objectives: Lonicera japonica Flos, widely used in traditional East Asian medicine, exhibits antipyretic, detoxifying, and anti-inflammatory activities and is rich in flavonoids, phenolic acids, and iridoids. Because the composition and yield of bioactive compounds vary with extraction conditions, this study aimed to evaluate the impact of extraction methods on the antioxidant potential of Lonicera japonica Flos. Methods: Powdered Lonicera japonica Flos was extracted using warm-water and hydrolysis-assisted methods. Antioxidant activity was assessed via DPPH and ABTS radical-scavenging assays. Major compounds were identified and quantified using UPLC–MS/MS, and molecular docking was performed to predict interactions between chlorogenic acid, isochlorogenic acid A, and the nuclear factor erythroid 2–related factor 2 (Nrf2) transcription factor. Results: UPLC–MS/MS analysis identified five major compounds in the warm-water extract and six in the hydrolyzed extract. The hydrolyzed extract showed a broader profile of caffeoylquinic acid derivatives, including cryptochlorogenic acid and 1,5-dicaffeoylquinic acid. Chlorogenic acid and isochlorogenic acid A were present at higher concentrations in the hydrolyzed extract (30.677 and 143.207 ppm, respectively) than in the warm-water extract. Both compounds exhibited significant DPPH and ABTS radical-scavenging activity from 10 μg/mL, with chlorogenic acid showing stronger effects. Docking analysis indicated stable binding to Nrf2 (–9.1 kcal/mol). Conclusions: Hydrolysis-assisted extraction enhances the recovery of antioxidant compounds from Lonicera japonica Flos, supporting its potential for standardized functional applications. Further studies are needed to confirm Nrf2 activation in biological systems.
