Development of Recessive Genotype for P34, 7S α′ Subunit, and Stachyose Components in Black Soybean with Green Cotyledon

초록

Black soybean cultivars or germplasms with green cotyledons have long been widely cultivated in Asia due to their high content of health-beneficial anthocyanins and lutein. However, mature seeds contain anti-nutritional and allergenic components such as P34, 7S α′ subunit proteins, and stachyose. Therefore, there is a need to develop black soybean genotypes without these components. A total of 126 F2 seeds with green cotyledons and black seed coats were obtained by crossing the female parent (p34p34 Cgy1Cgy1rs2rs2 genotype) with the male parent (p34p34cgy1cgy1Rs2Rs2 genotype). Thirty-five F2 seeds lacking the 7S α′ subunit protein were selected and grown into 32 plants in the field. Ten F2 plants with the rs2rs2 genotype were selected based on the Rs2 gene, a marker for stachyose content, using a DNA marker. The elite plant was selected based on plant type, maturity group (determined by maturity date), medium stem height, seed weight and seed condition. The selected plant had a maturity date of October 24, when sown on June 15, in the field. Its stem height was 55 cm, its 100-seed weight was 24.5 g, and its seed coat color and cotyledon color were black and green, respectively, for mature seeds. The selected plant has the p34p34cgy1cgy1 rs2rs2 genotype, which lacks P34 and 7S α′ subunit proteins, and has low stachyose content. It is expected to serve as a parent for breeding various colored soybean cultivars carring recessive genotypes for components of P34, 7S α′ subunit, and stachyose components, known to be anti-nutritional and allergenic.
