두 가지 DNA 마커를 이용한 MAS 기법을 통한 렉틴 및 P34 단백질에 대한 lelep34p34 유전자형 콩 선발

초록
Soybeans are a globally cultivated crop that serves as a crucial source of vegetable oil and protein. However, mature soybean seeds contain antinutrients, such as lectin and P34 proteins, which can cause indigestion and allergies. Marker-assisted selection using DNA markers to identify genotypes lacking these proteins at an early growth stage is highly effective for plant breeding. This study aimed to select individuals with the lelep34p34 genotype-which lacks both lectin and P34-using two DNA markers. A total of 108 F2 plants were obtained by crossing a female parent (LeLep34p34) and a male parent (leleP34P34). Using a DNA marker specific for the le allele, 22 F2 plants with the lele genotype were selected. Among these, eight were further selected based on evaluations of agronomic traits, including plant type, stem length, and lodging resistance. Using a second DNA marker specific for the p34 allele, four F2 plants with the p34p34 genotype were identified. After harvest, one plant with a yellow seed coat and a yellow hilum was finally selected. This plant matured around October 20, with a stem height of approximately 65 cm, a 100-seed weight of approximately 12.5 g, and a yield per plant of approximately 150 g. Notably, both lectin and P34 proteins were absent in the resulting F3 seeds. This selected plant is expected to serve as a valuable parent for breeding small-seeded yellow soybean cultivars lacking anti- nutritional and allergenic factors.
