Growth Responses of Cucumber Seedlings to Single and Combined Stresses of Mechanical Stimulation and Salinity

Earlier studies have reported that mechanical stimulation can enhance plant resistance to various environmental stresses. In this study, we evaluated the response of cucumber seedlings to salt stress combined with mechanical stimulation. Cucumber seeds were sown in 40-cell plug trays filled with commercial seedling medium. Brushing treatment using an acrylic film was applied every 2 hours, starting 29 days after sowing. After 7 days of brushing, NaCl solutions at concentrations of 50, 100, 150, and 200 mM were applied to both brushing and non-brushing groups. Growth and stress responses were measured 9 days after NaCl treatment. Plant height, leaf length, leaf width, leaf area, and fresh weights of shoots and roots decreased as NaCl concentration increased. However, SPAD values increased with higher NaCl levels. Brushing treatment reduced plant height regardless of NaCl concentration. While total root length and root volume decreased with increasing NaCl, brushing treatment mitigated these effects by increasing root length and root volume. MDA and H O levels increased with NaCl treatment, and the brushing group 22 showed higher contents of both than the non-brushing group. These results suggest that both mechanical and salt stresses increase stress responses in cucumber seedlings, and mechanical stimulation may alleviate salt-induced root growth inhibition. Therefore, it is suggested that stress tolerance may be induced through the regulation of the intensity of appropriate mechanical stimulation.
