Abstract
Objectives : The search for natural antioxidants that are effective against diseases induced by oxidative stress and that possess no side effects is currently being actively pursued. The primary aim of this study is to explore the biologically active compounds in the alcoholic extract of Inula helenium (IHAE), analyzed by LC-MS/MS, and to elucidate its antioxidant efficacy. Methods : Liquid chromatography-tandem mass spectrometry (LC-MS/MS) analysis was conducted using the UPLC system and the 3200 QTrap tandem mass system in positive ion mode at a wavelength of 284 nm. The free radical scavenging activity of IHAE was measured using the ABTS assay at wavelengths of 734 nm, respectively. To quantify the antioxidant compounds, the Folin-Ciocalteu method utilizing gallic acid was employed to determine the polyphenol content, while flavonoids were quantified using a colorimetric method with quercetin as the standard. Results : The analysis of six compounds extracted from IHAE using UPLC-MS/MS identified Glutathione, guanosine, 3-Indoleacrylic acid, Chlorogenic acid, Abscisic acid, and Marmesin as the principal precursor ions. In the ABTS assay, significant antioxidant activity was observed starting from a concentration of 10 μg/mL. The total phenolic content of IHAE was determined to be 11.07 ± 0.57 mg GAE/L, and the total flavonoid content was measured at 1.30 ± 0.47 mg QE/L. Conclusions : Based on these results, it has been confirmed that the six components contained in IHAE may possess strong antioxidant properties depending on the conditions. These findings can serve as a foundational basis for evaluating the antioxidant effects of IHAE and the potential of its active ingredients.
