dc.description.abstractObjectives: The overproduction and accumulation of free radicals and reactive oxygen species (ROS) result in oxidative stress at the cellular level, which play a critical role in the progression of numerous diseases. Plant-derived phenolic compounds are acknowledged for their substantial antioxidant activities, functioning as natural antioxidants within the organism. Therefore, the aim of this study was to identify bioactive compounds in the Lithospermum erythrorhizon alcoholic extract (LEAE) and to evaluate its antioxidant effect. Methods: Liquid chromatography-tandem mass spectrometry (LC-MS/MS) was used to identify the active compounds in the LEAE that may contribute to its antioxidant effects. The antioxidant effect of LEAE was evaluated using DPPH (2,2-diphenyl-1-picrylhydrazyl) and ABTS (2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)) assays. To further assess the antioxidant capacity, the total phenolic and flavonoid contents were also measured. Results: LC-MS/MS analysis identified six predominant compounds in LEAE: Adenosine, Guanosine, Salicylic acid, Lithospermic acid, Salvianolic acid B and Salvianolic acid A. In the DPPH assay, significant free radical scavenging activity was observed starting from an LEAE concentration of 60 μg/mL. Similarly, the ABTS assay demonstrated a concentration-dependent reduction in free radical levels, beginning at an LEAE concentration of 20 μg/mL. Conclusions: LEAE exhibits a significant antioxidant effect, which is attributed to the presence of adenosine, guanosine, salicylic acid, lithospermic acid, salvianolic acid B, and salvianolic acid A. These findings suggest that LEAE may serve as a significant source of natural antioxidants and possess promise for treating disorders associated with oxidative stress.-
